May 16
Amplified OxyS, SoxS, KatG, and HemH promoters and cleaned up the products with ethanol.
May 17
Restriction digested the products from the previous day.
May 18
Froze down bacterial strains: pGLOW, pPLAT, and pPLAT w/ GFP. Ligated above promoters into pPLAT w/ GFP and transformed them into DH5a E. Coli.
May 19
Prepared colony PCR of above transformed bacteria to ensure the ligation and transformation worked.
