iGEM team Amsterdam 2011
Welcome to the wiki of team iGEM Amsterdam 2011! On this site, you can find out everything about the team that represents Amsterdam in this year's edition of the iGEM competition, as well as our project: icE. coli. You will find answers to questions like...- Who are the team members?
- What are they working on?
- Who's paying for all this?
Please take a look around, and be sure to contact us with any questions you are left with!
Abstract
Escherichia coli's optimal growth temperature is 37°C. When decreasing the temperature the growth rate of the bacterium decreases rapidly. Once it reaches 7 °C, growth of wild type 'E. coli is impossible. In our project, we will try to increase the cold tolerance of E. coli, allowing it to grow faster at temperatures below 37°C. At the end of our project, we expect to be able to culture our bacteria at very low temperatures, possibly even at or below freezing point.
The approach we use involves heterologous expression of proteins in the DH5α E. coli strain. Out of different (combinations of) synthesized genes and parts submitted by previous iGEM teams, we will create cold resistance BioBricks – also known as CryoBricks.
These CryoBricks will be valuable to both fundamental and applied research. One of the many applications is replacing antibiotic resistance with cold resistance as a selection property in plasmid backbones. They can also be useful in various projects of other iGEM teams, and may open new avenues for heterologous expression of heat-sensitive proteins.
