- sequencing failed for all of P2SDM1, P2SDM2, P2SDM3.
- restriction digested the miniprepped plasmids to see if the plasmids get cut (if the site is still there, then the vector should cut and give a single band).
- gel verification of the digests suggest that the site has been removed (show bands that are identical to the uncut template/original vector).
