Lab Journal
Monday 20th June - Attempted transformations of E.coli with biobricks for the first time. We used the Banana odour generator; red and green luciferase; and Epic biobricks. We followed the protocol offered by the New England Biolabs for this procedure (see methods). We find out tomorrow if it worked!
Tuesday 21st June- The control cells and 'Epic' Biobrick cells were successfully transformed. The cells transformed with the 'Banana odour' Biobrick mysteriously vanished from the incubator and were found a day later in the chill room. All other transformations failed. We repeated protocols of all unsuccessful transformations.
Wednesday 22nd June- All transformations successful. Much relief. Made cultures from colonies of all transformations with the aim of making glycerol stock solutions as soon as possible.
Thursday 23rd June - Made glycerol stock solutions of all the successfully transformed E. coli cells from yesterday so that we will have an unlimited supply for the rest of the iGEM project.
