Double digest of ho1 and Cph8 set up and placed in 37 degree C incubator. Reaction mixture was as follows:

• 3 ul NEBuffer 3 (100% activity for both BglII and NotI)

• 1 ul BglII

• 1 ul NotI (acquired from Claudia's lab)

• 10 ul template DNA

• 16 ul ddH2O

Digestion should be run for 24 hours and quenched by heating at 65 degree C for 20 minutes.
