As we did for the tRNA we decided to synthesize the promoter Hyperspank ba y annealing extension with Klenow polymerase. That polymerase is known for elongating primers at 37°C that way we prevent the primers to detach when the product is heated to reach the elongation temperature of the Taq or phusion.

I launched one control and three tubes.

So it seems that 3 clones of tetR:YFP (2.2.1 - 2.2.2 - 2.3.1) and 2 clones of tetO array (4.7.1 -4.8.2) works.
Sent it today to sequencing.
