Monday 25 July

We spend the whole day planning our next step, given that the results call for a different approach. Designing primers, evaluating assembly methods... We leave some inoculums so we can start over with the mini-preps and all that stuff tomorrow.

What a tragedy! Our wonderful clock has fallen off the wall with a loud crash and it has smashed to pieces. RIP, dear pink clock.

Tuesday 26 July

Minipreps, digestions, etc. After using the NanoDrop to measure the DNA contained in our mini-preprs we come to the conclusion that this machine is absolutely useless. Nothing like the classical electrophoresis to check whether you've isolated plasmids properly!

Wednesday 27 July

XGal and IPTG have come in a box with 4 kg of dry ice. You know what that means?? A 15-MINUTE-PARTY IN THE LAB! (and a broken glass as a result of an experiment gone wrong, but don't panic, nobody's been hurt). Erlenmeyers, graduates and glass bottles have also arrived today; very delayed, but better late than never!

Thursday 28 July

Today's Rocío's birthday!Our "little" girl is 21 years old, so she ows us a beer for a start. After the lab we'll have a bit of a do.

[Everything else that happened this day obviously lacks interest when compared to this important event, but anyway, we'll let you know one more trivial detail: we've already transformed our ligations!]

Friday 29 July

The party was a complete success yesterday (Rocío got lots of presents), so today nobody's properly awake yet. Even so, we're able to go on discussing what we'll have to do next week. It seems our ligations are ok, at least there's something in the dishes, but they're too few colonies, so we're not sure whether they're correct ligations or some other kind of life.
