Purpose:
Transformation of ligation mixture. Used TOP10 chemical mixture and electrocompetent cells.
Procedures:
-Standard heat shock transformation for the TOP10 cells
-Standard electrocompetent transformation but one transformation was n-butanol purified while the other was straight from the ligation mixture.
Notes:
The n-butanol purification is to remove the ligation salts to prevent arcing during the electric shock.
