1. Gel verification for CASO (1-8)
Observation: in second row, GFP spcer, tetR spacer, CRISPR#1, and CRISPR #2
2. Gel verification by digestion
3. Purify tetR from Gel
4. Ligate pSB1C3 with tetR, place in the 15c incubator overnight
5. Transformation

1. Transform ligations of tetR in DH5α and top10 competent cells
2. Inoculate PCOLA, PCDF and CRISPR
07/22/2011
1. Mini-prep for :
tetR in DH5α and top10 (16 samples)
CRISPR (2 samples)
PCOLA (1 sample)
PCDF (5 samples)
2. Digest tetR with EcoR1 and Pst1
3. Test the digested tetR by running a gel
