7/16/2011	All but negative control grew 12.2.13 is slightly pink probably due to the constant red Glycerol stock- 350 µL of cells and 150 µL if 50% glyrcerol Miniprep and transformation results- the plate had about 50 to 60 colonies

7/17/2011	Incubate in 3 mL LB broth overnight at 37 degrees Celsius and 2020 rpm

7/18/2011	Colony grew, negative control didn’t Transfomation- 50 µL DH5a, 2 µL DNA at 37 degrees Celsius overnight (all carb) Glycerol stock- 350 µL of cells and 150 µL of 50% glycerol Miniprep Dissolving and diluting primers (10 pmol/µL) PCR and gel electrophoresis and purification Digestion for 1 hour and 30 minutes at 37 degrees Celsius Gel Electrophoresis and Purification Digestion and ligation

7/19/2011	Transformation- 50 µL DH5a, 2 µL DNA at 37 degrees Celsius overnight
