03/August/2011 • Sergio gave us some recommendations about bacteria induction. • Min inoculum 13L, 8G, 2M, 22P, sac and 24C to have an stock of cells and minipreps of construct B • Inoculum of DH5-α, sac, pET12a to verify the restriction site of EcoRI. • IPTG induction of 6 different strains: C41, C43, BW27783, XLIBlue, BL21 and Rosseta gami. • We pick 2 colonies of each strain for overnight • Minipreps of yesterday’s stock • Digestion with EcoRI and PstI forward electrophoresis.
