Purpose:
Concentrate and determine if NotI is exhibiting star activity.
Procedures:
For OmpA:
Tube 4- OmpA+2 enzymes with 2 hour incubation at 37 degree and heat inactivation then freezer.
Tube 5- OmpA+2 enzymes overnight at 37 degrees.
Tube 6- OmpA+ NdeI with 2 hour incubation at 37 degree then NotI added for overnight.
For pBAD:
Tube 1: Same conditions as Tube 5.
Tube 2: Same conditions as Tube 4.
Tube 3: Same conditions as Tube 6.
