Performed a gel extraction of pcyA and ho1 digest products. Concentration of pcyA was 22.2 ng/ul. Concentration of ho1 was 20.9 npg/ul.

Ligation reaction set up as follows:

• 2 ul ligase buffer

• 3 ul T4 DNA ligase

• 10 ul purified ho1 digest product

• 4.8 ul purified pcyA digest product

• 0.2 ul ddH2O

Ligation reaction to be incubated at room temperature for 24 hours.
