Today we had colonies on the plate with A2-2 in the expression vector and one colony on the plate with A1-A in pSB1C3. We will not do further with these colonies until monday morning.

Labwork

• Made new amp plates

• Yesterday we run a pcr on the A1-A in expression vector, A1-B in expression vector, A1-A in pSB1C3 and A2-1 in pSB1C3. Today we will test if the ligations were succesfull by running the pcr products on a gel.

The gel showed no bands at 4600 bp, which we had expected, so something must be wrong.
