Objective: obtain INP fragments
PCR reaction volumes
5 uL DNA template
5 uL forward primer
5 uL reverse primer
15 uL taq green mix
PCR conditions
95 degrees >> 4 min
95 degrees >> 30 sec
60 degrees >> 30 sec
72 degrees >> 1 min
34 cycles
72 degrees >> 10 min
- INP tube (~1kb)
- INP negative control (no DNA added)
- GFP tube (less than 1kb)
- GFP negative control (no DNA added)
Results:
Thick bright bands at 1kb (INP) and ~950bp(GFP)
No bands from the negative controls.
Conclusion: there is something wrong with vent polymerase.
