Building a Synchronized Oscillatory System

Synchroscillator: Data page/parts submitted to the registry

Parts Submitted to the Registry

[http://partsregistry.org/Part:BBa_K546002 BBa_K546002] - 3OC6HSL Receiver Device with a lux pR regulated GFP (including a LVA Tag) Reporter: AHL responsive element that produces a low constitutive level of luxR via lux pL. Combined with AHL, luxR induces lux pR. lux pR regulates the expression of GFP. Upon addition of AHL the GFP expression increases.

[http://partsregistry.org/Part:BBa_K546000 BBa_K546000] - Autoinducer 3OC6HSL Receiver Device with a lux pR Controlled Autoinducer Synthetase: AHL responsive element that produces a low constitutive level of luxR via lux pL. Combined with AHL, luxR induces lux pR. lux pR regulates the expression of autoinducer synthetase.

[http://partsregistry.org/Part:BBa_K546005 BBa_K546005] - Autoinducer 3OC6HSL Receiver Device with a lux pR Regulated GFP (including a LVA Tag) & Autoinducer Synthetase: AHL responsive element that produces a low constitutive level of luxR via lux pL. Combined with AHL, luxR induces lux pR. lux pR regulates the expression of autoinducer synthetase. The system creates a positive feedback loop, yielding high titres of GFP.

[http://partsregistry.org/Part:BBa_K546001 BBa_K546001] - 3OC6HSL Receiver Device with a regulated 3OC6HSL inactivation enzyme gene: AHL responsive element that produces a low constitutive level of luxR via pL. Combined with 3-oxohexanoyl-homoserine lactone, luxR induces pR. lux pR regulates the autoinducer inactivation enzyme A (aiiA) gene that degrades AHL.

[http://partsregistry.org/Part:BBa_K546546 BBa_K546546] - Tunable Synchronized Oscillatory System: This system mainly consists of a positive and negative feedback loop, that are both tunable. With the right parameters oscillations can be created when the loops are active, as there is an interplay between the loops. Both the positive and negative feedback loops are AHL responsive. Without tuning our team succeeded in creating oscillations.

Data for pre-existing parts

[http://partsregistry.org/Part:BBa_K546003 BBa_K546003] - 3OC6HSL Receiver Device with lux pR: 3-oxohexanoyl-homoserine lactone (AHL) responsive element that constitutively produces an intermediate level of LuxR via lux pL promoter. Combined with AHL, luxR activates lux pR promoter. This part is a repaired version of [http://partsregistry.org/Part:BBa_F2621 BBa_F2621] that appeared to be defective (R0063 missing), when we sequenced the part.
