Tuesday

We are two men down today. One with fewer and one with a sprained foot. But the sun is shining and the spirit is high!

Lab Work

• There were no colonies on the plates from yesterday. We checked all of our calculations and made some corrections.

• We did mutations again, with the corrections. Afterwards we digested our mutants with Dnp - see protocols Mutations

• Transformed XL1-Blue competent cells - see protocol Transformations

• We did a Mini prep on the A1 overnight culture from yesterday. We cut the plasmids with PstI-HF and ran a gel with the resulting DNA. Great Succes!!! Only one band appeared, indicating that we succesfully did a point mutation in the target PstI restriction site. The DNA has now been sent to sequencing and expect the answer in two or three days.
