Created TENS
- 4.5 mL TE and
- 250uL 10% SDS
- 250uL 2N NaOH
- -Spin 4 tubes (1.5mL) 30"
- -Pour off sol'n & resuspend
- -Add
- 300uL TENS: mix
- 150uL NaOH: mix
- -Spin 2.5 min.
- -Transfer supernatant to clean tube and add 1mL of EtOH
- -Mix and pellet DNA by centrifugation for 2-5 min
- -Wash pellet with 70% ethanol and allow pellet to dry
- -Resuspend the pellet in 30λ of the TE
- -Digest 5-10λ
