12/July/2011 • Minipreps -> 14L col.3, 8G col.3 and 13L8G col 19 • Digestion with EcoRI , SpeI and XbaI ; 2m with PstI and SpeI • Electrophoresis • Chema gave us certain strains for the expression of the protein, which are: Roset Gami, XL1 Blue, C43, Pet 12, BL21, C41, Pet 28 • Purification -> 13L col.3 and 8G col.3 • Ligation -> 13L+8G • Touchdown PCR for SacC, Program: 61-43ºC, Δ1ºC every 2 cycles. • PCR product purification, quantification and overnight ligation in pGEM.
