Materials and Equipment for iGEM.

Here is a list that helps you prepare for the iGEM competition. This list has the materials needed and the equipment that will be used in the iGEM competition. Some of the reagents are generic because the project will specify what type of reagent you will use. Also the list is divided if you have a thermocycler or not.

Lab with a Thermocycler

Yeast

This materials list is a beginning point for the lab. There are protocols for E. Coli and Yeast on openwetware that are helpful.

  • Growth media: YPD
  • Selective media: SD with amino acids added
  • Agar: YPD, Electrophoresis agar
  • Restriction enzymes: Biobrick kit from New England Science will help
  • Buffers: TAE/TBE/or LB
  • Seed DNA
  • Stains for gels
  • Enzymes: DNA and RNA Polymerase (if needed), ligase, protease
  • dNTP’s
  • Chemicals: MgCl, Tris-HCl
  • Competent cells both ecoli and yeast

Equipment for the lab: PCR, pressure cooker used as an autoclave, micropipettes and tips, hemocytometer or spectrophotometer, tubes, plates, incubator, water bath or shaking water bath.


E. Coli

This materials list is a beginning point for the lab. There are protocols for E. Coli and Yeast on openwetware that are helpful.

  • Growth Media: T broth, M9, LB
  • Selective Media: Antibiotics added to media
  • Agar: LB agar, Electrophoresis agar
  • Restriction enzymes: Biobrick kit from New England Science will help
  • Buffers: TAE/TBE/or LB
  • Stains for gels
  • Enzymes: DNA and RNA Polymerase (if needed), ligase, protease
  • dNTP’s
  • Chemicals: MgCl, Tris-HCl
