Digest of pcyA and ho1 set up as follows:
- pcyA digested with XbaI and SpeI
- ho1 digested with SpeI and PstI
ho1 digest product should be treated with phosphatase to prevent religation to self. pcyA digest should be gel purified and extracted to remove backbone. These two products should be ligated together, transformed and a colony PCR should verify which colonies contain the full construct.
