Created TENS

4.5 mL TE and

250uL 10% SDS

250uL 2N NaOH

-Spin 4 tubes (1.5mL) 30"

-Pour off sol'n & resuspend

-Add

300uL TENS: mix

150uL NaOH: mix

-Spin 2.5 min.

-Transfer supernatant to clean tube and add 1mL of EtOH

-Mix and pellet DNA by centrifugation for 2-5 min

-Wash pellet with 70% ethanol and allow pellet to dry

-Resuspend the pellet in 30λ of the TE

-Digest 5-10λ
