Dr Liz I'll be available for phone consulting from 2:00 - 6:00+ @ 443.610.5259. I'm very glad Patrick can open the lab today as I'm not feeling well (on antibiotic since yesterday) and have much paperwork still to finish re: iGEM's use of Smith 491. l will be working from home all day unless I begin to feel much better.

Steve Stowell I'll be in 2:30 to 3:00, leaving about 7:30, or a convenient stopping point.


Prepare two gels to be stored in 491 refrigerator

2% GelPilot LE Agarose

100 mL TAE 1X Buffer

2g GelPilot LE Agarose (1,000-23,000bp)


Prepare 10 uL PCR product with 2 uL 6x Blue loading dye to be stored in 491 freezer. These are the colony PCR products that were amplified on 8/18/11 from the transformations on 8/4/11.

Next lab samples need to be loaded into the gels and ran. Transformation of 5I and 5K showed no growth. These transformations should be redone next lab.
