Wednesday

We messed up again. We had no cultures on our plates and we don't know what went wrong. The explanation we cling to is that it is our competent cells that aren't competent enough. So we have borrowed some from a very talented lab person and keep our fingers crossed.

Lab work

Did a mini-prep of our Biobrick - see protocol Miniprep
Checked the size of purified plasmid on an agarose gel and saw that it corresponded with the size from iGEMs website.
Did transformation again.
