China's wine culture has developed for thousands of years,and the wine as a traditional drink is closely linked with people's life. But excessive drinking will bring a lot of inconvenience, causing many social problems. For example, in recent years, drunk driving has become a ferocioustraffic killer.Therefore,we hope to develop a kind of engineering bacteria, which can reproduct in the human body , and has the ability of decomposition of excess alcohol function.

As the host bacteria of exogenous gene expression,lactobacillus' superiority is mainly manifested in the following:1.lactobacillus can colonize in the respiratory system, digestive system and urinary system, playing an important role in maintain ecological balance; 2.lactobacillus has confirmed that has no pathogenicity in food industry's various fields of long-term application, is recognized as the security level (GRAS, generally recognized as safe) microorganisms.As a result , we choose the lactic acid bacteria as recipients in gene engineering.

In the human body 's metabolism process, the alcohol is mainly catalyzed into acetaldehyde acetaldehyde by alcohol dehydrogenase ( ADH) in liver, further by acetaldehyde dehydrogenase (ALDH ) catalytic transformation into acetic acid.We found two genes expressing of ADH and ALDH, ADH1B2 and ALDH2, hoping to put them into BioBrick, introducing into Lactobacillus expression. At the same time we will introduce promoter of lactate detection as well as self-cracking mechanism, improving the gene pathway.

Super E.coli Architect is a design software of engineering bacteria. It offers a convenient and efficient design platform,which contributes to making simple ideas into a complete solutions. The existing synthetic biology softwares involve some specific functions such as finding genes and metabolism pathways, modeling and biobrick design.And these softwares are often oriented to scientific researchers. But we think that thereis a hope in synthetic biology to turn the science of artificial biosystem into a engineering program having a standard process. Therefore, we constructed a software, Super E.coli Architect, oriented to designers more than scientists.Providing all the necessary technical support,SuperE.coli Architect can release desighners from technical details,helping them work as the architects,allowing them to take more attention into constructing kinds engineering bacterias for solving practical problems.
