Stress sensor (rrnB+LacI+pLac+mCherry)
The stress sensor is our main construct. It is made from the following biobricks:

The promotor is a variant of BBa_K112118, that we have made ourselves using PCR. The stress sensor has a total length of 2653 bp, and is located inside the plasmid pSB1A2, carrying ampicillin resistance. We have put the stress sensor through several tests (see figure)
We have discovered that growing the cells containing the biosensor in a medium low in amino acids is the main way to activate it. As the figure shows, growing the cells in temperatures higher than 37 °C has little effect compared to growing them in a minimal medium. Already as we started this project we knew that lack of amino acids was the main trigger of ppGpp. Therefore, our results seems to indicate that our stress sensor work the way it is supposed to.
