Purpose: to digest pUC-pcyA-ho1 and pUC-pTet-Cph8 with appropriate restriction enzymes to put pTet-Cph8 in pUC-pcyA-ho1. Procedure: The following digestion reactions were prepared:

• 16 ul ddH2O

• 1 ul SpeI

• 1 ul PstI

• 3 ul NE buffer 2

• 10 ul pUC-pcyA-ho1 miniprep product (139 ng/ul)

• 16 ul ddH2O

• 1 ul XbaI

• 1 ul PstI

• 3 ul NE buffer 2

• 10 ul pUC-pTet-Cph8 (105 ng/ul)

Digest reactions placed in 37 degree C incubator overnight. Results: None
