Problems arose with the cloning of the LovTAP and Not-Gate in pSB1C3 vector. A reason might be that the mutation of the SpeI restriction site to NehI wasn´t succesful and the promotor of the LovTAP containing SpeI restriction sites was digested. Other difficulties were the primer design for the PCR of the LovTAP for the gibson assembly.
