Our Parts Image

The hydrogen production pathway is composed of four proteins. Our hydrogenase is the translational fusion of the gene codiffying a ferredoxin and hydA. PFOR is a Pyruvate Ferredoxin Oxido-Reductase that helps with the electron flux. The other two proteins are maturases, i.e. enzymes required for the proper assembly of the hydrogenase. These genes are organized in two operons. The first one comprises the maturase and PFOR and is regulated by the Rizhobium etli promoter pNifH1. The second one is regulated by pLacZ and contains the maturases. Periplasm export tags were added to the hydrogenase and PFOR.

Codon optimization

Our systems final host is R. elti. Given this, we optimized our sequences for R. etli's codon usage.

Periplasm export tags

As a Gramm-negative bacterium, Rhizobium etli has a periplasmatic compartment. Proteins can be exported into it with the appropriate peptide signal.
