We have discovered that growing the cells containing the biosensor in a medium low in amino acids is the main way to activate it. As the figure shows, growing the cells in temperatures higher than 37 °C has little effect compared to growing them in a minimal medium.

Already as we started this project we knew that lack of amino acids was the main trigger of ppGpp. Therefore, our results seems to indicate that our stress sensor work the way it is supposed to.

The stress sensor is our main construct. It is made from the following biobricks:
