We had no cultures on our plates and we don't know what went wrong. The explantation we cling to is that it is our competent cells that aren't competent enough. So we have borrowed some from a very talented lab person and keep our fingers crossed.

Wednesday

We fucked up again. We had no cultures on our plates and we don't know what went wrong. The explanation we cling to is that it is our competent cells that aren't competent enough. So we have borrowed some from a very talented lab person and keep our fingers crossed.

Lab work

  • Did a mini-prep of our Biobrick - see protocol Miniprep
  • Checked the purified plasmid on an agarose gel.
  • Did transformation again.
