We autoclaving 1.5ml eppendorff tubes and then were treat at 800C for 2h in the microwave for nuclease elimination.
Orlando prepare chloramphenicol (100µg/ml)
We pass our Biobrick from Agar medium + chloramphenicol to LB medium + Chloramphenicol.
Sandra use as negative control : 200µl plasmid (RBS+T) in medium with Amp and inoculate in LB medium with Chlorampheenicol. No colonies have to grow because not have resistance for chloramphenicol.
Incubate at 370 C in shaker.
