We have discovered that growing the cells containing the biosensor in a medium low in amino acids is the main way to activate it. As the figure shows, growing the cells in temperatures higher than 37 °C has little effect compared to growing them in a minimal medium.

The stress sensor is our main construct. It is made from the following biobricks:

Biobrick Part number
LacI with RBS BBa_J24679
Terminator BBa_B0015
pLac BBa_R0011
mCherry with RBS and terminator BBa_J06702
