The platereader experiment worked well, showing that with enough ATC concentration RFP can be induced even with constitutive expression of TetR. The first graph shows the maximal RFP signal corresponding to each ATC dilution, averaged over the 4 different cell colonies. The second graph shows EFP expression over time for 2 individual wells, either without ATC or with 800 ng/mL.
