Butanol pathway >>
Data >>

Expected that the volume of kivd protein will be regulated by incubation temperature so that the production of isobutanol can be affected.

Isobutanol標準品的減量線:

DH5α is very suitable for this circuit, because it can provide higher production rate of Isobutanol.

When we additionally insert the following genes, Alss, ilvC, and ilvD, the production of Isobutanol will extremely increase. (About 80 times)

Diagram3.

Method: After we incubate the E.coli at 37 degree Celsius to OD 0.5, we will transfer them into different incubation degree, such as 30, 37, and 42 degree, then observe the production of isobutanol at 0 and 24 incubation hours.

Culturing in different temperature, it tends to that the production of isobutanol will increase with lower incubation temperature.

Method:

Utilizing our 2010 IGEM temperature controlled simulation, we incubate the E.coli at 37 degree Celsius to OD 0.5, we will transfer them into different incubation degree, such as 30, 37, and 42 degree, then observe the production of isobutanol at every single time. (Flow cytometry)

Under different temperature, there is a tendency that the production of isobutanol will increase with lower incubation temperature. Comparing the simulation of kivd protein production, decreasing temperature truly reduce the protein expression.
