• DNA digestion with SspI and NdeI + Buffer 3 at 37 C , 1 hr.
• Electrophoresis 60 V, 1.5 dr.
• Pre-inocolum of virgin cells in 15-20 microliters of LB without antibiotic for calcium competent cells.
• Purification of DNA of the agarose gel.
