we prepared:  500ml LB medium  Autoclaving LB medium  Glycerol 30% for preservation  Check the medium with plasmids, colonies grows well; negative control doesn’t grow after 22h on incubation.  We inoculate bacterias with plasmids P+R, R+T and with Rh in liquid LB + Amp. Incubate at 37oC in shaker.
