A Brief Approach to our Idea - Un Leve acercamiento a nuestra idea

Our objective is to generate a genetically-modified bacteria "detect and alert" system, particularly as a defense aid for coffee plantations against fungi. Bacteria are built such that they can detect chitin, an organic compund found in fungal cell walls, amplify this initial signal, and finally alert the plant by stimulating an early hypersensitive response against infection, while controlling their own population density. We strongly believe this biocontrol method may prove to become a powerful tool for farmers and crop workers everywhere. In addition, our contructed "detect and alert" biobricks could be useful to plentyful synthetic biology experiments and future iGEM teams.

Using the bacteria Escherichia coli DH5α, the system will use two or three differently-modified bacteria in the system.

The first bacteria will detect the presence of Chitin and will produce chitinase and a signal molecule to engage the respone of the second bacteria. The second bacteria will react to the signal and will start to produce a signal to activate the hipersensitive response of the plant and will generate a scent of peppermint that can tell the farmer when there is an invasion. Additionaly, the second or third bacteria will generate a reactive that will control the growth of all three bacterias, avoiding the risk of overloading the host with bacteria.

Nuestro objetivo es el de generar un sistema de "detección y alerta" mediante bacterias genéticamente modificadas, particularmente como control biológico de hongos en plantaciones de café. Se contruyen bacterias capaces de detectar quitina, un componente orgánico presente en las paredes celulares de los hongos, amplificar esta señal inicial y finalmente alertar a la planta, estimulando así una respuesta hipersensible en contra de la infección a tiempo, al mismo tiempo controlando su densidad poblacional. Creemos firmemente que este mecanismo de biocontrol puede servir como una herramienta poderosa para el agricultor. Adicionalmente, las partes (bricks) sintetizados pueden ser de utilidad para experimentos en biología sintética y futuros equipos iGEM.

Inicialmente las pruebas se realizarán a nivel de laboratorio, empleando como microorganismo modelo a Escherichia coli DH5α, sin embargo, se espera desarrollar finalmente un producto que pueda ser aplicado en campo.

Para este fin se emplearan inicialmente dos o tres bacterias, la primera de las cuales detectará la presencia de quitina y producirá quitinasa y una molécula señalizadora para activar la respuesta de la segunda bacteria, la cual regulará la señal de la primera bacteria y a la vez producirá ácido salicílico para darle una señal a la planta de la presencia de quitina. Adicionalmente, generará un olor a menta que luego podrá ser remplazado por un marcador que permita informar al agricultor. La segunda o tercera bacteria se encargará de secretar un compuesto que controle el crecimiento de las otras dos bacterias, evitando así el riesgo de infección de éstas a la planta!
