Plasmid Extraction

This protocol will purify plasmids from bacteria, though identification of plasmids will need to be confirmed by PCR.


Reagents

  1. HEPES (N-[2-hydroxyethyl] piperazine N'[2-ethane-sulfonic acid])
  2. Sorbitol
  3. Percoll
  4. PEG 6000
  5. Ficoll
  6. Bovine serum albumin (BSA)
  7. Isoascorbic acid
  8. Sodium pyrophosphate
  9. Glutathione
  10. MgCl2.6H2O
  11. MnCl2
  12. Sodium-EDTA

Equipment

  1. High-speed centrifuge and rotor
  2. Spectrophotometer for chlorophyll measurements
  3. Orbital shaker, light suitable for algal cell growth, and timer to control light settings
  4. 27-gauge * 0.5 inch stainless stell syringe needles
  5. B-D Multifit 10mL glass Leuer-lock syringe
  6. 15mL Corex centrifuge tubes
  7. Hemocytometer (cell counter)
  8. Fine (artist style) paint brush
