Human Practices - Legislation<br />
<br />
Colombian legislation: Transgenic organisms are unfortunately still mainly a taboo in our geographical region. This happens both because of a general public ignorance on the matter, as well as a lack of will and legislation by the governments. Proof of this is the lack of national biosafety measures in Venezuela, which automatically prohibits the commerce or planting of Living Modified Organisms (LMOs). Ecuador actually explicitly proscribed any activity related to genetically modified organisms in its new Constitution (2008), second chapter, article 15. In 1999, Peru created the Law No. 27104 that is supposed to control all activities regarding LMOs and in 2003 began to work in the development of a Biosafety Regulation. However, Peru has still yet to publish such a document because of internal issues.<br />
<br />
Colombia stands as a regional pioneer together with Brazil by participating in the Convention of Biological Diversity, as well as successfully ratifying and legislating biosecurity measures according to the Cartagena Protocol of Biosafety. As Colombians, we want to help our country develop these techniques further by abiding local legislation as well as becoming a technological vanguard through the application and divulgation of synthetic biology.<br />
<br />
What's more, a free trade agreement has recently been approved by presidents Barack Obama and Juan Manuel Santos between the United States of America and Colombia, which automatically lower customs duties of many agricultural and industrial products to zero, and automatically demands an increase of competitiveness for both countries, but more dramatically for Colombia. In this matter, we strongly believe innovative processing of our most important exportations are crucial to stand as a competitive economy, and this why we decided to engineer synthetic biology for the improvement of our coffee production.<br />

Regarding our current legislation in more detail:<br />

Through the ratification of the Cartagena Protocol of Biosafety with Law 720 of 2002, Colombia remains true as a listed party country in the Convention on Biological Diversity of 1992 that has three main objectives: 1. The conservation of biological diversity, 2. The sustainable use of the components of biological diversity, and 3. The fair and equitable sharing of the benefits arising out of the utilization of genetic resources. The Cartagena Protocol is basically an international agreement to ensure the safe handling, transport, and use of living modified organisms (LMOs) that result from modern biotechnology.<br />
<br />
Colombian expresident Álvaro Uribe ordained the decree 4525 of 2005 through the Ministry of Agriculture and Rural Development by which it delimits Law 740 of 2002. The decree first defines important terms that define our project as modern biotechnology (in vitro application of nucleic acids in live cells or organelles for the surpassing of the natural physiological barriers of the reproduction or recombination not traditionally used in reproduction and selection), emphasizes the importance of biosecurity in these kinds of projects, and determines the competent authorities which we have to inform about our biosafety techiniques, as well as project advances. In our case this turns out to be the Colombian Farming Institute (ICA), to which we must ask for authorization for research using LMOs.<br />
<br />
The decree also determines the National Technical Committee of Biosecurity for LMOs with agricultural, livestock, fishing, commercial forest plantations, and agroindustry: the Minister of Agriculture and Rural Development, the Minister for Social Protection, the Minister of the Environment, Housing, and Territorial Development, Colciencias (the Administrative Department of Science, Technology, and Innovation) Director, and the ICA manager. This is the committee in charge of evaluating project viability and biosafety. The Ministry for Social Protection, the National Institute for the Surveillance of Foods and Drugs (Invima, Colombia's FDA), the Ministry of Agriculture and Rural Development, ICA, and the Ministry of the Environment, Housing, and Territorial Development are in charge of the control and close following of authorized projects, as well as taking protective and disciplinary measures against malpractice.<br />
<br />

As a complement to our low income schools educational efforts, we decided to participate in Exposcience, asked our university's communication department to make us a divulgational [http://www/component/content/article/537-una-bacteria-heroe-de-los-cafetales video] for the general public, and were selected by the most important local newspaper "El TIempo" that wrote an [http://www.eltiempo.com/vida-de-hoy/educacion/estudiantes-de-los-andes-clasificaron-al-mundial-de-biologia-sintetica_10595064-4 article] about us!<br />
<br />
ExpoScience is Colombia's largest science and technology fair where state of the art national and international advances are shown. Additionally, over 900 students are invited to come by the government, since it's also the gathering of the country's best universities with science programs - where they are given a place to impress and attract students. This year, we participated from the 18th to the 23rd of October with our own stand, where we were able to illustrate people from all ages what the genetic code is, how it works, introduce them to the concept of synthetic biology and introduce the iGEM competition to over 500 thousand fair visitors.<br />
<br />
More specifically, we prepared three activities meant for the general public in which we presented the basic mechanisms of gene transcription and translation, synthetic biology based on standard parts and its potential for our country through our project. Since we had students from almost every grade, we prepared educational activities as an empirical introduction to the aforementioned concepts.<br />
<br />
1. We made up a game where children could play while learning the mechanics of transcription and translation, deciphering the genetic code and building up proteins. The game consists of:<br />
• A game mat that represents a cell with a space or the nucleus and a strand of mRNA attached to a ribosome.<br />
• An inflatable pool.<br />
• Acrylic nucleotides with shapes that complement each other as they do in reality (G-C, U-A).<br />
• Acrylic aminoacids in different colors.<br />
Children were explained the basics of the code and made to get into the pool previously filled with the nucleotides and aminoacids to complete the code printed in the mat. Every three nucleotides they could decide which aminoacid they'd like to attach. The code was made in such a way that there was only one open reading frame possible and that there was a codon that repeated itself so participants had to use the same aminoacid twice.<br />
After the activity was over, we made a short lecture explaining them that this is the exact way cells operate, and that proteins ultimately determine the cell's function, thus the importance of the genetic code.<br />
<br />
2. We also designed second game where students were able to apply the basic concepts of synthetic biology using standard parts by constructing their own DNA sequence.<br />
<br />
The game consisted of iGEM's biobrick convention shaped cardboard pieces with an electrical circuit attached in such a way that a LED would turn on only if the different parts were put next to each other in an appropriate order. The parts are:<br />
• 4 promoters, 3 looking right and one looking left with different voltages.<br />
• 4 ribosome binding sites with different resistors.<br />
• 4 coding sequences with different LED colors.<br />
• 4 terminators.<br />
<br />
The different pieces were engineered in such a way that the expression of a coding sequence would be illustrated by the turning on of its LED. However, to add customizability we designed our different parts in these ways:<br />
• Promoters have the batteries that power up the circuit, as well as different resistors to illustrate how some promoters could express more or less of a given coding sequence (resulting in different levels of LED brightness).<br />
• Ribosome binding sites also contained different resistors so that coupled with different promoters, even more levels of brightness may be achieved.<br />
• Terminators would close the circuit so that in their absence no LED would turn on.<br />
• The connections between parts were made up of small pieces of metallic sponges attached to the flanking wires so that changing parts and connecting them became very easy.<br />
• Parts were also designed in such a way that ribosome binding sites could only be connected between a promoter and a coding through specific distances of the connecting sponges. In the same way, coding sequences could only be connected between a ribosome binding site and a terminator.<br />
<br />
A small introduction about how synthetic biology allows us to write and engineer our own code was made and volunteers were asked to try and connect the different pieces until the LED turned on. Once it finally did, we asked them to try different promoters, coding sequences, and ribosome binding sites and to tell us what happened. The volunteers were asked to return to the public and we then gave a short lecture about part interchangeability and the main functions of each of the parts. We at iGEM Colombia took Human Practices very seriously because we believe it to be one of the competition’s most important aspects. Our principle objective is that of generating educational material available for future iGEM teams. We've also designed simple activities that may help illustrate key synthetic biology concepts such as how the genetic code works and the basic logical order DNA assembly should carry. As a multidisciplinary group, we decided to look at our outreach programs from multiple perspectives:<br />

'''Education''': as Universidad de los Andes students we’re in a very privileged position compared to most of our fellow Colombians, since most of our population does not continue into higher education programs. With this in mind we researched the Colombia Ministry of Education and National Administrative Department of Statistics (DANE) data and publications in order to try and find possible causes for this fact. Through this, we realized that natural and basic sciences are not one of our students top career choices, easily surpassed by engineering and economics:<br />

It is disappointing to see how little interest science awakes in Colombia, especially in a country where only a million of the 12 million students enrolled in elementary, middle, and high school, enter universities.<br />
<br />
We also found out how students with low scores in the Icfes test (Colombian SATs) have undergraduate level studies desertion rates as high as 60% at the end of their fourth year. This we believe to be very worrying given the fact that most of these students graduated from low-income schools.<br />

To tackle the aforementioned issues, we got in touch with Alianza Educativa (Educational Alliance), a non-profit organization whose purpose is to bring high level education to low-income schools. They helped us get in touch with several elementary, middle, and high schools for which we prepared a keynote presentation and Q&A section to try and motivate students into continuing their studies into higher education, particularly in science. Our lectures were basically an overview of biology through multiple perspectives, beginning with classical biology, where we made a short historical introduction to the study of life and metabolism. We then continued into cell biology, where we explained how organisms were composed of cells, and how they were the basic functional unit of multicellular beings such as ourselves. Next, we explained molecular biology as the fusion of genetics and biochemistry, contrasting genes in chromosomes as books in a library containing recipes for the production of proteins, and these as nanoscopic machinery that power up cells. Once you understand the importance of the genetic code for life, you may begin to question how these genes and proteins interact and generate genetic networks, and this is what systems biology is all about. Here, we emphasized how biology is not just for biologists, and how biological questions are in need of multidisciplinary teams to come up with interesting and more robust answers. Finally, we introduced students to the concept of synthetic biology, the engineering of new biological functions through the use of standard biological parts, the iGEM spirit, and a small module trying to change the common perception that bacteria and microbes in general are evil through past competition’s projects as examples and our own project as a means towards the improvement of a national export product as important as coffee. The Q&A section was an opportunity we gave students to come up personally and ask us anything they wanted regarding science and iGEM. We were very pleased with the quality of the questions and how receptive and curious students were about possible projects for synthetic biology. This is as far as we went before the Americas' Jamboree. However, we had still lots of interesting ideas we've tried to carry out before the World Finals!<br />

In order to make our experience and presentation materials a useful tool for other groups, we designed a questionnaire to be given to a subgroup of the children that attended our lectures (~150) and a group of control students of the same school who did not attend (~50). This feedback was used to redesign our presentations to better achieve our objectives. <br />
<br />
Our objectives for the educational activities can be summarized in three points:<br />
- Lack of information about synthetic biology and mistrust of genetic technologies, in particular, modified bacteria.<br />
- Interest in science, its usefulness and perceived difficulty of a career in science.<br />
- Specific knowledge on molecular, systems and synthetic biology.<br />
<br />
Our questionnaire tried to address these points with few questions that would be relevant in levels 7-12. The results can be summarized as follows (sample graphs illustrate percentage of each group per answer): <br />

Here we had a definite success in increasing exposure and interest in Synthetic Biology and changing perceptions about the inherent dangers of Living Modified Organisms (LMOs). Students had a more nuanced concept of genetic modifications and bacteria, as shown also by an absolute decrease in students that chose "bacteria are a recipe for disease" in a complete-the-sentence question. Our conclusion here is that we achieve our objective but need to prepare materials that can be given to the students after the activities to help them learn more by themselves.<br />

Here we had a moderate success improving interest, but what we found was that interest in science and positive perception of its practice and usefulness were much higher than the governmental information had led us to believe (showing a low rate of entrance to higher education in fields related with science) . To investigate this discrepancy further, we need to do a similar survey in rural schools, because we concentrated on low income schools in an urban setting. With respect to perceived usefulness of science we also saw a small change, so we will leverage the possible utility to farmers of our project, and we will explain their plight and the ecological consequences of fungicide abuse in our next version. Other groups would also have to use examples of applied iGEM projects.<br />

Here again we had a very satisfying outcome, as we saw that not only a notable increase in their perception of their knowledge of molecular biology after a single activity, but also in simple test questions about the basic concepts of molecular synthetic, and systems biology there was a clear improvement. <br />

'''Education''': as Universidad de los Andes students we’re in a very privileged position compared to most of our fellow Colombians, since most of our population does not continue into higher education programs. With this in mind we researched the Colombia Ministry of Education and National Administrative Department of Statistics (DANE) data and publications in order to try and find possible causes for this fact. Through this, we realized that natural and basic sciences are not one of our students top career choices, easily surpassed by engineering and economics:<br />

It is disappointing to see how little interest science awakes in Colombia, especially in a country where only a million of the 12 million students enrolled in elementary, middle, and high school, enter universities.<br />

We also found out how students with low scores in the Icfes test (Colombian SATs) have undergraduate level studies desertion rates as high as 60% at the end of their fourth year. This we believe to be very worrying given the fact that most of these students graduated from low-income schools.<br />

To tackle the aforementioned issues, we got in touch with Alianza Educativa (Educational Alliance), a non-profit organization whose purpose is to bring high level education to low-income schools. They helped us get in touch with several elementary, middle, and high schools for which we prepared a keynote presentation and Q&A section to try and motivate students into continuing their studies into higher education, particularly in science. Our lectures were basically an overview of biology through multiple perspectives, beginning with classical biology, where we made a short historical introduction to the study of life and metabolism. We then continued into cell biology, where we explained how organisms were composed of cells, and how they were the basic functional unit of multicellular beings such as ourselves. Next, we explained molecular biology as the fusion of genetics and biochemistry, contrasting genes in chromosomes as books in a library containing recipes for the production of proteins, and these as nanoscopic machinery that power up cells. Once you understand the importance of the genetic code for life, you may begin to question how these genes and proteins interact and generate genetic networks, and this is what systems biology is all about. Here, we emphasized how biology is not just for biologists, and how biological questions are in need of multidisciplinary teams to come up with interesting and more robust answers. Finally, we introduced students to the concept of synthetic biology, the engineering of new biological functions through the use of standard biological parts, the iGEM spirit, and a small module trying to change the common perception that bacteria and microbes in general are evil through past competition’s projects as examples and our own project as a means towards the improvement of a national export product as important as coffee. The Q&A section was an opportunity we gave students to come up personally and ask us anything they wanted regarding science and iGEM. We were very pleased with the quality of the questions and how receptive and curious students were about possible projects for synthetic biology. This is as far as we went before the Americas' Jamboree. However, we had still lots of interesting ideas we've tried to carry out before the World Finals!<br />

In order to make our experience and presentation materials a useful tool for other groups, we designed a questionnaire to be given to a subgroup of the children that attended our lectures (~150) and a group of control students of the same school who did not attend (~50). This feedback was used to redesign our presentations to better achieve our objectives. <br />
<br />
Our objectives for the educational activities can be summarized in three points:<br />
- Lack of information about synthetic biology and mistrust of genetic technologies, in particular, modified bacteria.<br />
- Interest in science, its usefulness and perceived difficulty of a career in science.<br />
- Specific knowledge on molecular, systems and synthetic biology.<br />
<br />
Our questionnaire tried to address these points with few questions that would be relevant in levels 7-12. The results can be summarized as follows (sample graphs illustrate percentage of each group per answer): <br />

Here we had a definite success in increasing exposure and interest in Synthetic Biology and changing perceptions about the inherent dangers of Living Modified Organisms (LMOs). Students had a more nuanced concept of genetic modifications and bacteria, as shown also by an absolute decrease in students that chose "bacteria are a recipe for disease" in a complete-the-sentence question. Our conclusion here is that we achieve our objective but need to prepare materials that can be given to the students after the activities to help them learn more by themselves.<br />

Here we had a moderate success improving interest, but what we found was that interest in science and positive perception of its practice and usefulness were much higher than the governmental information had led us to believe (showing a low rate of entrance to higher education in fields related with science) . To investigate this discrepancy further, we need to do a similar survey in rural schools, because we concentrated on low income schools in an urban setting. With respect to perceived usefulness of science we also saw a small change, so we will leverage the possible utility to farmers of our project, and we will explain their plight and the ecological consequences of fungicide abuse in our next version. Other groups would also have to use examples of applied iGEM projects.<br />

Here again we had a very satisfying outcome, as we saw that not only a notable increase in their perception of their knowledge of molecular biology after a single activity, but also in simple test questions about the basic concepts of molecular synthetic, and systems biology there was a clear improvement. <br />
We have yet to implement a feedback system on our hands-on educational materials, but we also want to use some feedback to generate improved versions that will be useful to other groups in the future.<br />

Our objective was to analyze the of our system for chitin detection and activation of the chininolytic metabolism of Vibrio fisherii ES144, we proposed two different strategies:<br />
First, we needed to characterize the expression of the two constitutive promoters of the two-component system (Sensor Promoter and CBP Promoter). We made a construct where the CBP promoter leads the expression of the mCherry reporter. The characterization was made using fluorometry and using as control the constitutive promoter of E. coli (BBa_J23119).<br />

The characterization of the chitin sensor on E. coli will be made on the future using the same strategy.<br />

Aditionally, we needed to made the characterization of the inducible promoter of the two component system (ChiP) as a response of the extracellular chitin concentration. In order to do that we realized a construct where ChiP was Upstream from the mCherry reporter. Without a termination codon, mCherry expression is controlled by the ChiP promoter.<br />
This construct will then be introduced into bacteria that contain plasmids that have the sequence for the Sensor and for CBP, where the level of fluorescence will be a measure of the promoter activation inducible to different extracellular concentrations of chitin. For this characterization the vectors have already been constructed and the transformation will be done soon. We´ll be sure to show you our results at the Jamboree!!<br />
