Thursday

It looks like we might have our first transformed colonies. Later today we hope to be able to pick single colonies for overnight cultures. We have purified the mysterius colony we found yesterday and we'll get it sequenced soon. And we have found a reason for our poor results with the mutated CYP transformations. It looks like a mathematical error mysteriously have made us use 100x the primer amount needed. We are therefore mutating for the third time - at least we get really good at it.

Lab work

  • Did miniprep of XL1-Blue transformed with CYPA2 - see protocol Mini prep
  • Mutated our CYP - see Protocol Mutations
