Purpose:
To try another method of transformation using electrocompetent cells instead of the chemical competent cells.
Procedures:
-Thaw competent cells in ice, also place cuvettes in ice.
-Add 1 uL of ligation into the thawed competent cells
-Incubate in ice for 5 minutes.
-Shock with electric current
-Add SOC or LB liquid to cuvette
-Plate
Results:
Negative= 0 colonies
OmpA Ligation= 0 colonies.
