Purpose: to isolate pUC-pcyA-ho1 plasmid from 4 3ml overnight cultures.
Procedure: used Promega Wizard Plus miniprep kit according to manufacturer specifications.
Results:
Colony 1 has a concentration of 139.3 ng/ul
Colony 2 has a concentration of 122.5 ng/ul
Colony 3 has a concentration of 57.2 ng/ul
Colony 4 has a concentration of 118.3 ng/ul
Using Colony 1 from amp plate 1 and Colony 4 from plate 2 for digest.
