Set up a PCR reaction to amplify ompR promoter from distribution plates (BBa_R0082). Fifty ul reaction volume contained:

• 1 ul template DNA

• 1 ul forward primer

• 1 ul reverse primer

• 5 ul thermopol buffer

• 0.5 ul 20 mM dNTPs

• 2.5 ul 10 mM MgSO4

• 0.5 ul Vent polymerase

• 38.5 ul of H2O

Silicatein alpha amplified from TOPO-Silicatein using the same reaction mixture as above.
