Daisy is working with the synthase 3-carene; 3-carene has three restriction sites that need to be modified (EcoRI, XbaI, PstI). She worked over the weekend, and has one more site to modify.

We learned that the truncated version of the synthase will increase the efficiency of the enzyme.

Daisy did a multiple site-directed mutagenesis using three sets of forward and reverse primers which contain the desired changes.

Sunday

Daisy did a DpnI digest of her multiple site-directed mutagenesis product and ran it on a gel. The appropriate sizes were shown on the gel, running at 7.6 kb. Daisy transformed the products into Dh5 alpha cells.
